چکیده

In the course of our antifungal screening programme, we have devised an efficient screening system for chitin synthesis inhibitors. Considering this as a therapeutic target, the fungal cell wall has a promising unique layer that fulfills the criteria for selective toxicity1^ The major structural cell wall components of many medically important yeasts, filamentous fungi, and dimorphic fungi consist mainly of glucans and chitin2). It is also known that complete depletion of chitin synthesis is lethal to fungi. Thus we have taken a wild type Saccharomyces cerevisiae (strain SS 553) and its chitin synthetase I-

A modified screen for the detection of cell wall-acting antifungal compounds.

One would logically expect that antifungal agents which act via the inhibition of cell wall biosynthesis should show fungicidal activity and low toxicity as the cell wall is essential for fungal cell growth and is not synthesized by mammalian cells. A variety of cell wall-acting antifungal agents have been discovered and some of these have become economically important1~3). However, new nontoxi...

Utilization of target-specific, hypersensitive strains of Saccharomyces cerevisiae to determine the mode of action of antifungal compounds.

Target-specific hypersusceptible strains of Saccharomyces cerevisiae were used to screen antifungal compounds. Two novel Erg7p inhibitors were identified, providing proof of principle of the approach taken. However, observed hypersensitivities to antifungals acting via other targets imply that use of this tool to identify the mode of action requires significant deconvolution.

Overexpression of chimeric chitinase42 enhanced antifungal activity of Trichoderma harzianum against Fusarium graminearum

Evidence for the role of chitinases in biocontrol by Trichoderma species has been well documented.Chit42 lacks a chitin–binding domain (ChBD) which is involved in its binding activity to insoluble chitin. The objective of the present study was to enhance antifungal activity of T. harzianum by overexpression of wild type and hybrid forms of Chit42 containing chitin binding domain. To produce chi...

New Benzimidazoles Derivatives: Synthesis, Characterization and Antifungal Activities

One of the most important goals in medicinal chemistry is the development of new heterocyclic compounds with pharmaceutical activity. Thus, a novel series of the derivatives of benzimidazole were synthesized and the structures of all the synthesized compounds have been confirmed by IR, 1 H- and 13C-NMR, Mass Spectroscopy and elemental analysis. The title compounds have been evaluated for antifu...

Biological Evaluation of Heterocycle Moiety of Some Novel azoles Derivatives as Antibacterial and Antifungal potential Agents

Background & Objective: Azole nucleuses are very important part of antimicrobial, analgesic and anti-inflammatory drugs. The azole class of compounds is the most popular among the antibacterial and antifungal classes because of its lower toxicity, higher efficacy and a broad spectrum of activity. Today, Efforts have focused on the development of new, less toxic and more efficacious antifungal a...
